Organic Matrix Derived from Host–Microbe Interplay Contributes to Pathological Renal Biomineralization

Matrix stones are a rare form of kidney stones. They feature high percentage hydrogel-like organic matter, and their formation is closely associated with urinary tract infections. Herein, comprehensive materials biochemical approaches were taken to map the organic–inorganic interface gather insights into host–microbe interplay in pathological renal biomineralization. Surgically extracted soft slimy matrix examined using micro-X-ray computed tomography various microspectroscopy techniques. Higher-mineral-density laminae positive for calcium-bound Alizarin red. Lower-mineral-density revealed periodic acid-Schiff-positive filamentous networks varied thickness. These networks, which featured polysaccharide content, enriched zinc, carbon, sulfur elements. Neutrophil extracellular traps (NETs) along immune response-related proteins, including calprotectin, myeloperoxidase, CD63, CD86, also identified networks. Expressions NETs upregulation polysaccharide-rich mucin secretion proposed as part host defense “trap” pathogens. host−microbe derived matrices can facilitate heterogeneous nucleation precipitation inorganic particulates, resulting macroscale aggregates known “matrix stones”. plausible aggregation constituents through underscore unique “double-edged sword” effect response pathogens biominerals.
